Description:Ponceau S Protein Staining Solution is a ready-to-use membrane stain for evaluating the transfer efficiency of a western blot.
Directions for Use:This product is recommended for rapid and reversible protein staining on nitrocellulose or PVDF membranes. This staining technique is often utilized to confirm protein electrotransfer in Western blotting assays prior to antibody-based detection.
1. Following protein electrotransfer, rinse the membrane with deionized water to completely remove the transfer buffer.
2. Incubate membrane in Ponceau S Staining Solution for 5-15 min at room temperature. The membrane background is currently red, and the protein bands are not visible or are indistinct.
3. Wash membrane with ddH2O until the protein bands become clearly visible.
4. Observe and preserve the staining results.
5. Wash membrane in 1X TBST several times for 5 min at room temperature until protein bands are no longer visible.
6. Proceed with blocking step of Western blot procedure.Precautions for Use:1. This product is not suitable for protein detection on nylon membranes.
2. PVDF membranes must be activated with methanol again after the protein electrotransfer.
3. Some components in the electrophoresis buffer (e.g., SDS) can interfere with the binding of Ponceau S to proteins. The membrane requires multiple washes.Storage/Stability:Store at 4°C for one year.
